Group Sequential Methods with Applications to Clinical Trials

Group Sequential Methods with Applications to Clinical Trials by Christopher Jennison and Bruce W. Turnbull presents a comprehensive presentation of group sequential methods. Written by active researchers in this area, it provides an ideal source for those wishing an introduction to the area and for those who desire a clear outline of specific topics or methods.

Group sequential methods answer the needs of clinical trial monitoring committees who must assess the data available at an interim analysis. These interim results may provide grounds for terminating the study-effectively reducing costs-or may benefit the general patient population by allowing early dissemination of its findings. Group sequential methods provide a means to balance the ethical and financial advantages of stopping a study early against the risk of an incorrect conclusion.

This text describes group sequential stopping rules designed to reduce average study length and control Type I and II error probabilities. The authors present one-sided and two-sided tests, introduce several families of group sequential tests, and explain how to choose the most appropriate test and interim analysis schedule. Their topics include placebo-controlled randomized trials, bio-equivalence testing, crossover and longitudinal studies, and linear and generalized linear models.

Research in group sequential analysis has progressed rapidly over the past 20 years. This text surveys and extends current methods for planning and conducting interim analyses. It provides straightforward descriptions of group sequential hypothesis tests in a form suited for direct application to a wide variety of clinical trials. Medical statisticians engaged in any investigations planned with interim analyses will find this book a useful and important tool.

It should quickly become a standard reference both for those wishing to apply the methods and for researchers in the area. The extent of coverage of various topics broadly reflects current usage. The discussion of each topic appears to be well balanced. All in all, this is a very welcome book. The statistical methodology for sequential trials is complicated.

This book provides an excellent presentation of it. It succeeds as both a training text and as a reference source. I liked the progression of the book. Any statistician involved in designing or analyzing sequential trials should have a copy.

Drug Design: Structure- and Ligand-Based Approaches

Drug Design: Structure- and Ligand-Based Approaches by Kenneth M. Merz, Dagmar Ringe and Charles H. Reynolds provides a complete snapshot of the field of computer-aided drug design and associated experimental approaches. Structure-based (SBDD) and ligand-based (LBDD) drug design are extremely important and active areas of research in both the academic and commercial realms.

Methods utilized in computer-aided drug design can be classified into two major categories: structure based and ligand based, using information on the structure of the protein or on the biological and physicochemical properties of bound ligands, respectively. In recent years there has been a trend towards integrating these two methods in order to enhance the reliability and efficiency of computer-aided drug-design approaches by combining information from both the ligand and the protein.

Topics covered include X-ray crystallography, NMR, fragment-based drug design, free energy methods, docking and scoring, linear-scaling quantum calculations, QSAR, pharmacophore methods, computational ADME-Tox, and drug discovery case studies. A variety of authors from academic and commercial institutions all over the world have contributed to this book, which is illustrated with more than 200 images.

Authors also describe pseudoreceptor methods, pharmacophore methods, fingerprint methods and approaches integrating docking with similarity-based methods. In this article, we will describe the concepts behind each method and selected applications.

This is the only book to cover the subject of structure and ligand-based drug design, and it provides the most up-to-date information on a wide range of topics for the practicing computational chemist, medicinal chemist, or structural biologist.

Kenneth M. Merz received his PhD in organic chemistry at the University of Texas at Austin and completed postdoctoral research at Cornell University and the University of California, San Francisco. He is a member of the Quantum Theory Project and Professor of Chemistry at the University of Florida, Gainesville.

Foye's Principles of Medicinal Chemistry 7th Edition

Foye's Principles of Medicinal Chemistry 7th Edition by David A. Williams offers updated chapters plus new material that meets the needs of today's medicinal chemistry courses. This latest edition offers an unparalleled presentation of drug discovery and pharmacodynamic agents, integrating principles of medicinal chemistry with pharmacology, pharmacokinetics, and clinical pharmacy.

This text has been fully revised and updated to meet the changing curricula of medicinal chemistry courses. Emphasis is on patient-focused pharmaceutical care and on the pharmacist as a therapeutic consultant, rather than a chemist.

A new disease state management section explains appropriate therapeutic options for asthma, chronic obstructive pulmonary disease, and men's and women's health problems. Also new to this edition: Clinical Significance boxes, Drug Lists at the beginning of appropriate chapters, and an eight-page color insert with detailed illustrations of drug structures. Case studies from previous editions and answers to this edition's case studies are available online at the Point.

All the chapters have been written by an international team of respected researchers and academicians. Careful editing ensures thoroughness, a consistent style and format, and easy navigation throughout the text. This is still the gold standard against which all other medicinal chemistry books must be judged. It is content rich, yet attempts to relate the discipline to modern clinical practice. It is comprehensive and accessible, and an excellent resource for students as well as professionals seeking to update their knowledge.

The addition of the clinical significance sections alone justifies replacing the previous edition. Following an introduction to drug discovery and its relation to absorption, metabolism, and physiochemical and biopharmaceutical properties of drugs, each of the book's subsequent three parts addresses particular drug classes.

The division of subjects is rational and informative. Of particular note is the last section on disease state management with particularly unique coverage of women's and men's health. It is here that students can see the clinical application of a diverse group, such as gonadal steroids. The clinical vignettes and clinical significance explanations throughout the book are unique and important. The book's main shortcoming (not for me, however), as with many detailed tomes, is its sheer size.
